It means any 3rd party tools such as alternative apps, or any program that does something on Twitter will have to pay or stop working.
An API is just a set of instructions on how to make your software work with whatever the API is for.
So if I wanted to make a new app that takes the days most googled phrase and searches it on Twitter and finds the most popular tweet, I wouldn’t be allowed to do that without paying.
