The real changes occur at the post-synaptic membrane. There is a down-regulation of receptor sites in the presence of excess neurotransmitter. It takes a few weeks for this to occur and for the body to achieve homeostasis.

No. The transmitters don’t degrade that quickly after they are used and reabsorbed.

Without the drugs, the transmitters are released, used, and excess is reabsorbed. On the drugs, the reabsorption is delayed to that they have more time to trigger the next nerve – nothing about their composition changes.
