How do we find which compounds alter our state of mind?

I understand the traditional “test it and find out method.” But I mean aside from introducing the compounds/chemicals into the body, how else do people find out what can be considered a drug? Did someone just come across 2CB and thought it sounded cool and took some and increased the dose until they were “high?” Is there a specific molecular structure that’s sought out? How?

You do indeed begin with the tried and tested drugs, which have had their chemical structures identified. These have certain functional groups – such as the OH in alcohol. It’s a reasonable assumption that if C2H5-OH does strange things to your brain, than maybe CH3-OH will too. Of course, in the case of ethanol and methanol one is a drug and the other a poison, but the principle is sound.
