There’s not one single process that can describe all tolerance. Generally speaking the body likes to be in equilibrium and has a lot of built in negative feedback loops to try and maintain that equilibrium. For instance many drugs work by activating certain receptors on cells. If those receptors are constantly being activated then the cells may start to express less of those receptors. This creates a tolerance for the drug abd also accounts for certain withdrawal symptoms because stopping the drug will no longer return you to equilibrium.. it will just throw you out if balance in the other direction.
