At least where I live, if I make a transfer between my accounts (with different banks) too late on a Friday, the money won’t be available until Monday. But there are no humans involved in this process – it seems to me that could be instantaneous as far as the technology is concerned. What am I missing?

They could if they wanted to. Some comments mention that people are needed to support those automated systems. Those people already work on weekends. It’s not like a bank turns off all computers on Friday evening and turns them back on on Monday. Those servers run 365 days a year. It actually is programmed not to send your money over the weekend. So it’s extra programming to check for days of week and holidays.
