There is a fine line between anesthesia and murder drug. That’s why anesthesiologists have so many years of training and make so much money.

You can’t just inject a random amount of drug into a person and get a perfect result. It would be too easy to accidentally kill someone.

Tranquilizers need to be measured to the size of the person you are using them on. If you use too much, you’ll kill them. If you don’t use enough, it won’t actually be effective. As such, using them like they are used in movies to instantly knock out people isn’t really practical, and not much research into them has been done as a result.

To add onto the answers already here, such a drug just doesn’t exist. Even under the best, most controlled circumstances (like actual surgery, where a perfect dose can be calculated for the patient, the patient is generally calm and not in fight-or-flight, and you can safely use IV lines to administer agents as directly as possible) it will take a few seconds.
