eli5: what will happen if you have pain(like broken bone) and you don’t take pain killers? how body and brain will react/adjust?

Long story short, the body will release a group of hormones called endorphins. They are secreted from the hypothalamus and pituitary gland, and will increase in concentration in your blood once your body detects that pain signal (ie a broken arm). The endorphins essentially block the pain sensation in your brain, however this sensation only lasts temporarily. Pharmaceutical painkillers mimic this effect and through chemistry are actually more potent and sensational than the body’s natural painkillers.
